目的研究混合菌在大鼠根尖周炎过程中对Toll样受体2(Toll-like receptors 2,TLR2)的表达影响。方法取20只SD大鼠,随机分为4组,每组5只。对鼠的右侧下颌第一磨牙开髓,A1~A4组封药时间分别为1、2、3和4周。对照为健康的左侧下颌第一磨牙。按照实验设计的时间点处死SD大鼠,提取右下颌第一磨牙根尖区组织,采用Real-time PCR检测TLR2mRNA的表达情况。结果对照中有少量TLR2mRNA表达,A1和A2组TLR2mRNA的表达水平(1.71±0.10 vs 2.49±0.22)呈增高趋势,分别是对照数值(1.00)的1.71倍和2.49倍(P<0.05);A3组最高峰(2.94±0.14)为对照(1.00)的2.94倍(P<0.05);但A4组(1.79±0.14)开始下降。A2与A3两组(2.49±0.22 vs 2.94±0.14)比较差异有统计学意义。结论 TLR2在大鼠的混合菌感染的根尖周炎全过程中均有所表达,其表达水平在根尖周炎中发挥着重要作用。
Objective To study the effect of mixed bacteria on the expression of Toll-like receptors 2 (TLR2) during periapical periodontitis in rats. Methods Twenty SD rats were randomly divided into 4 groups with 5 rats in each group. The first right molar mandibular first molars opened, A1 ~ A4 group sealing time were 1, 2, 3 and 4 weeks. Control for a healthy left mandibular first molars. SD rats were sacrificed at the experimental design time points, and the tissue of apical region of the first mandibular first molar was extracted. Real-time PCR was used to detect the expression of TLR2 mRNA. Results There was a small amount of TLR2 mRNA expression in control group. The expression levels of TLR2 mRNA in A1 and A2 groups (1.71 ± 0.10 vs 2.49 ± 0.22) were increased, which were 1.71 and 2.49 times higher than those in control (1.00) (P <0.05) The highest peak (2.94 ± 0.14) was 2.94 times that of the control (1.00) (P <0.05), but the value of A4 (1.79 ± 0.14) began to decline. A2 and A3 two groups (2.49 ± 0.22 vs 2.94 ± 0.14) difference was statistically significant. Conclusion TLR2 is expressed in the whole process of mixed periapical infection in rats. The expression level of TLR2 plays an important role in periapical periodontitis.
