目的:研究影响栀子提取物黏膜吸收的有关因素。方法:以离体蛙皮为模型进行体外黏膜渗透实验,研究渗透介质的渗透压和pH对栀子提取物指标成分栀子苷表观渗透系数Papp的影响,考察蛙皮对栀子苷稳定性影响。结果:以生理盐水(pH 6.88)、纯水、1.8%NaCl溶液、生理盐水(pH 4.05)、生理盐水(pH 10.05)作为渗透介质,栀子苷蛙皮渗透Papp值分别为(0.53±0.01),(0.21±0.05),(0.44±0.12),(0.42±0.13),(0.26±0.03)cm.min-1,以生理盐水为渗透介质12 h的累计渗透率可达(55.69±9.81)%,无明显时滞。在蛙皮作用下,栀子苷8 h左右开始降解,降解速率常数K=1.999,半衰期为0.347 h。结论:栀子提取物黏膜渗透性良好,符合零级吸收过程,渗透压及pH对渗透影响显著,符合生理环境的等渗偏中性介质更有利于栀子苷的渗透吸收,蛙皮对栀子苷降解作用不影响黏膜渗透研究,离体蛙皮模型可较好研究栀子提取物的黏膜渗透过程。

Conclusion: The extract of musk extract has good mucous membrane permeability, which accords with the zero-order absorption process. The osmotic pressure and pH have a significant effect on the penetration. The isotonic neutral medium that accords with the physiological environment is more conducive to the penetration and absorption of geniposide. The degradation of glucosides did not affect the study of mucosal penetration, and the isolated frog skin model could better study the mucosal infiltration process of wolfberry extract.
