目的:通过HPLC法定量测定中药材暗紫贝母在不同组培时期的贝母甲素含量。方法:采用CosmosilODS2C18(5μm,250mm×4.6mm),以水—乙腈—二乙胺(体积比:70:30:0.03)为流动相,流速为1.0ml·min-1。结果:贝母甲素在1~10μg与峰面积的对数值呈线性关系,各批次回收率较好,且其各RSD值稳定。结论:本实验能够较好测定暗紫贝母在不同组培时期贝母甲素的动态积累量,并能确定其最佳积累时期。
Objective: To quantitatively determine the content of Fritillaria cirrhosa in different tissue culture stages by HPLC. Methods: Cosmosil ODS2C18 (5μm, 250mm × 4.6mm) was used as the mobile phase at a flow rate of 1.0ml · min-1 with water-acetonitrile-diethyl amine (70: 30: 0.03). Results: Fritillaria in 1 ~ 10μg and the peak area of the logarithmic value was linear, the recovery rate of each batch is good, and its RSD values ?? stable. Conclusion: This experiment can better determine the dynamic accumulation of Fritillaria platyphylla in different tissue culture stages, and determine the optimal accumulation period.
