目的:探讨活化STAT3蛋白抑制蛋白(protein inhibitor of activated STAT3,PIAS3)在不同分级分期的人膀胱癌及正常膀胱上皮中的表达情况及其临床意义。方法:利用SP三步法免疫组织化学技术检测PIAS3在膀胱癌及正常膀胱上皮中的表达。结果:PIAS3在正常膀胱上皮的表达率和表达水平均显著低于膀胱癌组织(P=0.0000002;P=0.000003);且表达水平随分化程度的降低而升高(P=0.00001);在浸润性膀胱癌(T2~T4)中的表达水平显著高于其在表浅膀胱癌(Tis~T1)中的表达(P=0.02)。结论:试验结果提示,PIAS3蛋白异常表达可能与膀胱癌的发生发展关系密切;它可能成为具有潜在价值的膀胱癌标志物或者基因治疗新靶点。
Objective: To investigate the expression of PIAS3 in human bladder cancer and normal bladder epithelium with different grading stages and its clinical significance. Methods: SP three-step immunohistochemistry was used to detect the expression of PIAS3 in bladder cancer and normal bladder epithelium. Results: The expression and expression level of PIAS3 in normal bladder epithelium were significantly lower than those in bladder cancer (P = 0.0000002; P = 0.000003). The expression level of PIAS3 increased with the differentiation degree (P = 0.00001) The expression level of bladder transitional cell carcinoma (T2 ~ T4) was significantly higher than that in bladder cancer (Tis ~ T1) (P = 0.02). Conclusion: The results suggest that the abnormal expression of PIAS3 protein may be closely related with the occurrence and development of bladder cancer. It may become a potential target of bladder cancer or a new target of gene therapy.
