目的探讨奥沙利铂对人结肠癌细胞(SW620)核转录因子κB(NF-κB)、信号传导与转录激活因子3(STAT3)表达影响。方法体外培养SW620细胞至对数生长期后,设不同浓度奥沙利铂(40、80、160μg/m L)干预组,同时设对照组,72 h后,应用四甲基偶氮唑盐法检测奥沙利铂对SW620细胞的增殖抑制率;应用Hoechst 33258细胞核染色法观察细胞形态改变;同时应用蛋白印迹法(Western blot)检测细胞内源性NF-κB、STAT3蛋白表达。结果与对照组比较,奥沙利铂低、中、高剂量组SW620细胞增殖抑制率[分别为(66.19±5.93)%、(86.92±7.85)%、(96.85±8.86)%]升高(P<0.05),细胞凋亡数[分别为(9.46±1.05),(18.57±2.16),(27.29±3.48)个/视野]上升(P<0.01);与对照组比较,奥沙利铂低、中、高剂量组SW620细胞内源性NF-κB及STAT3蛋白表达量[分别为(0.674±0.016)、(0.435±0.012)、(0.212±0.009)和(0.485±0.015)、(0.264±0.008)、(0.113±0.004)]明显下降(P<0.01)。结论奥沙利铂能够抑制SW620细胞增殖,其机制可能与抑制SW620细胞内源性NF-κB、STAT3表达有关。
Objective To investigate the effects of oxaliplatin on the expression of nuclear factor κB (NF-κB), signal transducers and activators of transcription 3 (STAT3) in human colon cancer cell line SW620. Methods After SW620 cells were cultured in vitro to logarithmic growth phase, different concentrations of oxaliplatin (40, 80, 160 μg / mL) were given to the intervention group. At the same time, the control group was treated with tetramethylzirconate The inhibitory rate of oxaliplatin on the proliferation of SW620 cells was detected. The morphological changes of cells were observed by Hoechst 33258 nuclear staining. Meanwhile, the expression of NF-κB and STAT3 were detected by Western blot. Results Compared with the control group, the proliferation inhibition rate of SW620 cells in low, medium and high dose oxaliplatin groups was (66.19 ± 5.93)%, (86.92 ± 7.85)% and (96.85 ± 8.86)%, respectively (P0.05), and the number of apoptotic cells (9.46 ± 1.05, (18.57 ± 2.16) and (27.29 ± 3.48) / visual fields, respectively) increased in comparison with the control group (0.674 ± 0.016), (0.435 ± 0.012), (0.212 ± 0.009) and (0.485 ± 0.015) and (0.264 ± 0.008), respectively, in medium and high dose groups of SW620 cells , (0.113 ± 0.004)] decreased significantly (P <0.01). Conclusion Oxaliplatin can inhibit the proliferation of SW620 cells, which may be related to the inhibition of endogenous NF-κB and STAT3 expression in SW620 cells.
