目的:研究肿瘤基因Caspase-3与PTHrP和Sox9在人体软骨肉瘤细胞HTB-94中的相互作用机制。方法:小干扰siRNA分别阻断PTHrP和Sox9后,检测肿瘤细胞中Caspase-3的mRNA和蛋白表达。结果:PTHrP基因被干扰后,Caspase-3的mRNA表达增加了57.7%,蛋白的表达增加了57.1%;Sox9基因被干扰之后,Caspase-3的mRNA表达增加了47.4%,蛋白的表达增加了48.2%。结论:肿瘤基因Caspase-3在软骨肉瘤细胞中的表达与软骨肉瘤相关基因PTHrP和Sox9有关,可能是位于PTHrP和Sox9的下游基因。
OBJECTIVE: To study the mechanism of the interaction between tumor suppressor gene Caspase-3, PTHrP and Sox9 in human chondrosarcoma cell line HTB-94. Methods: Small interfering siRNAs blocked the expression of Caspase-3 mRNA and protein in tumor cells after blocking PTHrP and Sox9 respectively. Results: The mRNA expression of Caspase-3 was increased by 57.7% and the expression of protein was increased by 57.1% after PTHrP gene was disrupted. The mRNA expression of Caspase-3 increased by 47.4% and protein expression increased by 48.2% after Sox9 gene was disrupted %. Conclusion: The expression of Caspase-3 in chondrosarcoma cells is related to chondrosarcoma-related genes PTHrP and Sox9, which may be downstream genes of PTHrP and Sox9.
