为探讨N一甲基一D一天冬氨酸(NMDA)受体在c一fos基因表达及其与癫痫易感性的关系,我们以Northern印迹杂交法测定结果显示:(1)外源性NMDA可诱导脑细胞c一fos基因的mRNA表达;竞争性或非竞争性NMDA受体拮抗剂可完中阻断c一fos基因mRNA表达。(2)NMDA诱导脑内c一fos的mRNA表达随神经元发育呈上升趋势,于体外培养24天达高岭。(3)在发育中,听源性癫痫易感大鼠脑细胞的c一fos基因mRNA表达均高于对照组,且在细胞培养18天时差异有显著意义。提示NMDA受体活性的变化可能与癫痫易感性的形成有关。
To investigate the relationship between N-methyl-D-aspartate (NMDA) receptor expression in c-fos and its association with epilepsy susceptibility, we used Northern blot hybridization assay to show that: (1) Exogenous NMDA Induced c-fos gene mRNA expression in brain cells; competitive or non-competitive NMDA receptor antagonist can block c-fos gene mRNA expression. (2) NMDA-induced c-fos mRNA expression in brain increased with the development of neurons, cultured for 24 days in vitro. (3) During the development, the mRNA expression of c-fos gene in brain cells of susceptible epilepsy rats was higher than that of the control group, and the difference was significant at 18 days of cell culture. Suggesting that changes in NMDA receptor activity may be related to the formation of epilepsy susceptibility.
