目的研究内皮素(ET)-1在慢性心力衰竭大鼠心肌的表达及生长激素释放肽(GHRP)的干预作用,并探讨其相关机制。方法 60只雄性Wistar大鼠随机分为对照组、模型组和GHRP组,每组20只。采用腹腔注射阿霉素法建立心衰模型;检测心功能变化;HE染色观察心肌细胞形态变化;ELISA法检测心肌组织匀浆中ET-1含量。结果心功能和心肌细胞形态与对照组比较,模型组有显著变化,而GHRP组变化不明显。心肌组织中ET-1的含量,模型组高于对照组和GHRP组(P<0.05),而GHRP组和对照组间无明显差异(P>0.05)。结论 GHRP能降低心肌组织中ET-1的表达,具有保护和改善心功能的作用。
Objective To investigate the effect of endothelin (ET) -1 on myocardium expression and growth hormone releasing peptide (GHRP) in rats with chronic heart failure, and to explore its possible mechanism. Methods Sixty male Wistar rats were randomly divided into control group, model group and GHRP group, with 20 rats in each group. The model of heart failure was established by intraperitoneal injection of doxorubicin. The changes of cardiac function were detected. The morphological changes of myocardial cells were observed by HE staining. The content of ET-1 in myocardial homogenate was detected by ELISA. Results Compared with the control group, cardiac function and cardiomyocyte morphology changed significantly in the model group, but not in the GHRP group. The content of ET-1 in myocardium was higher in model group than in control group and GHRP group (P <0.05), but there was no significant difference between GHRP group and control group (P> 0.05). Conclusions GHRP can reduce the expression of ET-1 in myocardium and protect and improve the cardiac function.
