目的对杜仲Eucommia ulmoides进行血清药物化学研究。方法采用UHPLC-Q-TOF-MS技术,通过比较杜仲提取物、给药组及空白组大鼠血清的指纹图谱,确定口服杜仲提取物后大鼠血中移行成分。结果给药组大鼠血清指纹图谱与空白组血清指纹图谱对比有明显不同。含药血清中出现7个移行成分,与杜仲提取物指纹图谱在相同保留时间出峰,为原型成分。采用对照品对照,确定5个色谱峰所表征的化学成分为原型吸收入血成分,依次为京尼平苷酸、原儿茶酸、京尼平苷、松脂醇二葡萄糖苷、松脂醇单葡萄糖苷。通过查阅文献,推断了2个色谱峰所表征的化学成分可能为1-羟基松脂醇二葡萄糖苷、杜仲醇。结论血中移行成分可能为杜仲的体内直接作用物质,为明确杜仲药效物质基础提供了科学依据。
Objective To study the serum drug chemistry of Eucommia ulmoides. Methods The fingerprint of serum of Eucommia ulmoides extract, administration group and blank group were compared by UHPLC-Q-TOF-MS technique to determine the migration components in the blood of rats after oral administration of Eucommia extract. Results Serum fingerprinting in the drug-treated group was significantly different from that in the blank group. There were 7 transitional components in the drug-containing serum, and the fingerprint of Eucommia extract peaked at the same retention time as the prototype component. Control samples were used to identify the five chromatographic peaks of the chemical composition of the prototype absorption into the blood components, followed by geniposidic acid, protocatechuic acid, geniposid, Pinoresinol glucoside, Pinoresinol monoglucose Glycosides. By consulting the literature, we conclude that the chemical components of the two peaks may be 1-hydroxynolide diglucoside and eugenol. Conclusion The blood migration components may be the direct role of Eucommia in vivo substances, to provide a clear scientific basis for the Eucommia pharmacological substances.
