目的 研究 3种砷化合物对健康人淋巴细胞DNA的损伤效应间是否存在作用差异。方法 无菌分离健康人静脉血淋巴细胞 ,分别染以亚砷酸钠 (AsⅢ)、砷酸钠 (AsV)及甲基砷酸钠(MAsV)。每种处理因素均分 5个剂量组 :1、5、10、2 0、5 0 μmol/L。培养 2 4h后进行单细胞凝胶电泳实验 (SCGE) ,计数彗星细胞个数 ,测量总彗星长度。结果 除MAsV 的 1μmol/L剂量组外 ,其他各因素各剂量组彗星细胞的频数分布与对照组相比差异均有显著性 (P <0 .0 5 )。除AsV1μmol/L和MAsV1、5 μmol/L 3个剂量组外 ,其他各因素各剂量组的总彗星长度均明显高于对照组。同一处理因素不同剂量组间以及不同处理因素相同剂量组间彗星细胞的频数分布及总彗星长度差异均有显著性 (P <0 .0 5 )。砷化合物作用剂量与彗星细胞比率及总彗星长度之间均具有相关性 (rAsⅢ =0 .8134,rAsV =0 .8734,rMAsV=0 .8994 )。结论 3种砷化合物均可引起健康人淋巴细胞DNA损伤。在同一处理因素作用下 ,砷化合物作用剂量与DNA损伤效应间存在剂量 -效应关系 ;不同处理因素同一作用剂量时 ,3种砷化合物对DNA的损伤效应间存在差异 ,损伤程度表现为AsⅢ >AsV>MAsV。
Objective To study the effect of three arsenic compounds on DNA damage in healthy human lymphocytes. Methods Aneurysmal blood lymphocytes were isolated aseptically and stained with AsⅢ, AsV and MAsV respectively. Each treatment was divided into 5 dose groups: 1,5,10,2 0,5 0 μmol / L. After cultured for 24 hours, single cell gel electrophoresis (SCGE) was performed, the number of comet cells was counted, and the total comet length was measured. Results The frequency distribution of comet cells in each dose group was significantly different from that in control group except for 1 μmol / L dose of MAsV (P <0.05). Except AsV1μmol / L and MAsV1,5μmol / L 3 dose groups, the total comet length of each dose group was significantly higher than that of the control group. The same treatment factors between different dose groups and the same dose of different treatment groups, the frequency distribution of comet cells and total comet length difference was significant (P <0.05). The dose of arsenic compound was related to the ratio of comet cells and total comet length (rAsⅢ = 0.83 4, rAsV = 0.8734, rMAsV = 0.9949). Conclusion All three arsenic compounds can cause DNA damage in healthy human lymphocytes. Under the same treatment factor, there was a dose-effect relationship between the doses of arsenic compounds and the DNA damage effects. The damage effects of three arsenic compounds on DNA were different at the same dosage of different treatment factors, and the damage degree was AsⅢ> AsV > MAsV.
