目的 为确定由PCR技术在小儿慢性分泌中耳炎中耳分泌液中检测到的细菌DNA是否来自于活的细菌。方法 观察中耳分泌液对DNA粘度和DNA酶消化DNA作用的影响。结果 中耳分泌液和DNA的粘度是稳定的 ,中耳分泌液不降解DNA ,DNA酶迅速消化DNA ,中耳分泌液显著抑制DNA酶消化DNA的活性。结论 应用PCR技术在大多数中耳分泌液中检测到的细菌DNA可能不是来自于活的细菌 ,可能是受到中耳分泌液保护的以前细菌感染的残余物
The purpose was to determine if the bacterial DNA detected by PCR in the middle ear secretions of chronic otitis media in children comes from live bacteria. Methods The effects of middle ear secretion on DNA viscosity and DNase digestion DNA were observed. Results The viscosity of middle ear secretions and DNA was stable. The middle ear secretions did not degrade DNA. DNase rapidly digested DNA. The middle ear secretions significantly inhibited the activity of DNase-digested DNA. Conclusions Bacterial DNA detected by PCR in most of the middle ear secretions may not be from live bacteria and may be residues of previous bacterial infections protected by middle ear secretions
