目的探讨不同质量分数高良姜素对人黑素瘤A375细胞黑素合成以及酪氨酸酶基因(TYR)、酪氨酸相关蛋白-1(TRP-1)和酪氨酸相关蛋白-2(TRP-2)mRNA的影响。方法选取人黑素瘤A375细胞,随机分为空白对照组、阳性对照组、90%高良姜素组和99%高良姜素组,其中阳性对照组、90%高良姜素组和99%高良姜素组各分为1.0,5.0和10.0 mmol/L组,每组给予相应浓度的含8-甲氧基补骨脂素、90%高良姜素和99%高良姜素的培养液,对照组给予等体积的培养液,采用MTT法测定细胞增殖情况,Na OH裂解法检测黑素合成情况,RT-PCR检测细胞TYR mRNA等表达情况。结果相同浓度下,90%高良姜素组和99%高良姜素组A值均高于阳性对照组(均P<0.05),且两组比较差异无统计学意义(P>0.05);10.0 mmol/L浓度下,90%高良姜素组和99%高良姜素组黑素水平分别为(170.12±0.07)%和(121.41±0.07)%,明显高于浓度为1.0 mmol/L和5.0 mmol/L浓度组(P<0.05);相同浓度下,90%高良姜素组黑素水平明显高于99%高良姜素组(P<0.05);在1.0 mmol/L浓度下,99%高良姜素组能显著上调TYP和TRP-2 mRNA表达,分别为(0.65±0.08)和(0.72±0.30),明显高于空白对照组以及相同浓度下90%高良姜素组(P<0.05);在5.0 mmol/L浓度下,99%高良姜素组能显著上调TYP,TRP-1,TRP-2 mRNA表达,分别为(0.61±0.07)、(0.61±0.11)和(0.67±0.27),明显高于空白对照组以及相同浓度下90%高良姜素组(P<0.05)。结论高良姜素对人黑素瘤A375细胞黑素合成及TYR,TRP-1,TRP-2 mRNA表达有所影响,其中99%高良姜素促进细胞黑素合成的能力弱于90%高良姜素,值得进一步研究。
Objective To investigate the effects of galangin with different mass fraction on melanin synthesis, tyrosinase gene (TYR), tyrosine-related protein-1 (TRP-1) and tyrosine-related protein-2 -2) mRNA.
