目的:优选石斛清咽含片的提取工艺。方法:采用HPLC测定石斛酚含量,检测波长278 nm,流动相乙腈(A)-0.5%甲酸(B)梯度洗脱(0~20 min,27%A;20~30 min,27%~40%A;30~45 min,40%~65%A;45~50 min,65%~27%A)。以石斛酚提取量和干膏率的综合评分为指标,在单因素试验基础上,通过Box-Behnken试验考察提取时间、料液比、浸泡时间对提取工艺的影响。结果:最佳提取工艺为加11倍量水浸泡35 min,提取3次,每次1 h;石斛酚提取量0.189 6 mg·g-1,干膏得率41.27%。结论:通过Box-Behnken效应面法及总评“归一值”为评价系统优选的提取工艺合理可行,为石斛清咽含片的应用提供参考。
Objective: To optimize the extraction process of Dendrobium Qingyan lozenge. Methods: The content of gigantol was determined by HPLC. The detection wavelength was 278 nm. The mobile phase was eluted with a gradient of acetonitrile-A (0.5% formic acid) (0-20 min, 27% A; 20-30 min, 27-40% A; 30-45 min, 40% -65% A; 45-50 min, 65% -27% A). Based on the single-factor test and the Box-Behnken test, the effects of extraction time, solid-liquid ratio and soaking time on the extraction process were evaluated by taking the comprehensive score of the amount of gigantol extracted and the dry matter rate as indexes. Results: The best extraction process was adding 11 times amount of water soaking 35 min, extraction 3 times, each time 1 h; The extract of gigantol 0.189 6 mg · g-1, dry paste yield 41.27%. Conclusion: Box-Behnken effect surface method and general comment “normalized value ” are reasonable and feasible for the optimal extraction system of the evaluation system, and provide a reference for the application of Dendrobium Qingyan Lube.
