目的 探讨血管内皮生长因子 (Vascularendothelialgrowthfactor,VEGF)及其受体 (VEGFR)KDR ,Flt - 1在急性白血病细胞株NB4、K56 2、HEL ,Jurkat、HL - 6 0 ,THP的基因表达情况 ,为进一步研究急性白血病治疗的抗肿瘤靶点提供思路。方法 采用RT -PCR和酶联免疫吸附法 ,对 6种急性白血病细胞系肿瘤细胞进行VEGF及其受体的基因型和表型检测。结果 6种白血病细胞系均 ( 1 0 0 % )检测到VEGF基因表达 ,强度不同 ,其中 3种细胞株 ( 50 % )检测到受体Flt- 1基因表达 ,所有细胞株均未检测到受体KDR基因表达 ;6种急性白血病细胞株上清液中VEGF浓度都明显高于对照。结论 VEGF及其受体在白血病细胞表达 ,说明VEGF及其受体在白血病的发生、发展中起重要作用 ,是急性白血病特征之一。
Objective To investigate the gene expression of vascular endothelial growth factor (VEGF), KDR and Flt - 1 in acute leukemia cell lines NB4, K56 2, HEL, Jurkat, HL - 60 and THP. To study the treatment of acute leukemia anti-tumor targets provide ideas. Methods RT-PCR and enzyme-linked immunosorbent assay were used to detect the genotypes and phenotypes of VEGF and its receptors in 6 acute leukemia cell lines. Results The expression of VEGF gene was detected in all six leukemia cell lines (100%) with different intensities. Among the three cell lines (50%), the expression of Flt-1 gene was detected, and no receptor was detected in all the cell lines KDR gene expression; 6 kinds of acute leukemia cell supernatant VEGF concentrations were significantly higher than the control. Conclusion VEGF and its receptor are expressed in leukemic cells, indicating that VEGF and its receptors play an important role in the development and progression of leukemia. It is one of the characteristics of acute leukemia.
