目的:研究清热益气中药黄连人参对药对自发性2型糖尿病大鼠胰岛细胞凋亡与炎症因子表达的影响。方法:采用高热量高脂饲料喂养诱导GK大鼠,建立自发性T2DM大鼠模型,予清热益气中药黄连人参对药进行干预4周,设Wistar大鼠作为对照,观察超敏C反应蛋白质、肿瘤坏死因子α水平及凋亡因子Bcl-2,Bax的表达情况。结果:模型组血清TNF-α显著高于正常组和治疗组(P<0.01),模型组和治疗组血清hs-CRP水平均显著高于正常组(P<0.01),模型组TNF-α在胰岛的表达水平显著高于正常组和治疗组(P<0.01,P<0.05),模型组Bax表达水平显著高于正常组和治疗组(P<0.01)。结论:清热益气中药黄连人参可降低糖尿病大鼠炎症因子水平,下调炎症因子及促凋亡因子的表达,抑制细胞凋亡,促进胰岛细胞损伤修复,也提示了2型糖尿病“壮火食气”的微观机制。
Objective: To study the effect of Chinese herb of Qingre Yiqi on the apoptosis of islet cells and the expression of inflammatory cytokines in spontaneously type 2 diabetic rats. Methods: GK rats were induced by high-calorie and high-fat diet and spontaneously T2DM rats were established. The rats were treated with Qingre Yiqi Chinese traditional medicine, Huanglian ginseng for 4 weeks. Wistar rats were used as control to observe the changes of hypersensitive C-reactive protein, Tumor necrosis factor α and the expression of Bcl-2 and Bax. Results: The level of serum TNF-α in model group was significantly higher than that in normal group and treatment group (P <0.01). The serum hs-CRP levels in model group and treatment group were significantly higher than those in normal group (P <0.01) The expression of Bax in model group was significantly higher than that in normal group and treatment group (P <0.01). The expression of Bax in model group was significantly higher than that in normal group and treatment group (P <0.01, P <0.05). Conclusion: Radix Ginseng ginseng can reduce the level of inflammatory cytokines in diabetic rats, down-regulate the expression of inflammatory factors and pro-apoptotic factors, inhibit apoptosis and promote the repair of islet cells, and also suggest that type 2 diabetes Gas "micro-mechanism.
