目的 :研究慢性束缚应激时大鼠下丘脑 β 内啡肽 (β endorpin ,β EP)的变化以及逍遥散、四君子汤、金匮肾气丸 3种中药复方对其的影响。方法 :用特制束缚架连续束缚 7天与 2 1天 ,每天 3h的方法 ,制作大鼠束缚应激模型 ,用免疫组织化学方法结合图像分析检测下丘脑β 内啡肽的变化。结果 :2 1天模型组较正常对照组大鼠下丘脑β EP免疫反应强度、阳性细胞数目及阳性细胞平均总面积均明显增强、增多、增大 (P <0 0 5 )。与 2 1天模型组比较 ,逍遥散组及肾气丸组能显著降低下丘脑 β EP免疫反应阳性强度 (P <0 0 5 ) ,减少下丘脑 β EP免疫反应阳性细胞数目 (P <0 0 5 ) ,减小下丘脑 β EP免疫反应阳性细胞平均总面积 (P <0 0 5 )。
Objective : To study the change of β-endorpin (β EP) in hypothalamus and the effects of three components of Xiaoyao San, Sijunzi Decoction and Jinkuishenqi Pills on chronic restraint stress. METHODS: The rat restraint stress model was established by using a special restraint frame for 7 days and 21 days, and 3 hours per day. The change of β-endorphin in the hypothalamus was detected by immunohistochemical method combined with image analysis. RESULTS: The β-EP immunoreactivity, the number of positive cells and the average area of positive cells in the hypothalamus on the 1st day of the model group were significantly increased, increased, and increased (P 0 05). Compared with the 2-day model group, Xiaoyao San and Shenqi Pills can significantly reduce the positive intensity of β-EP immunoreactivity (P <0 05) and reduce the number of β-EP immunoreactive cells in the hypothalamus (P <0. 5) Reduce the average total area of beta-EP immunoreactive cells in the hypothalamus (P < 0.05).
