目的探讨腺病毒携带肝细胞生长因子基因(Ad-HGF)不同途径转染对大鼠缺血心肌的影响。方法将75只大鼠随机分为心肌内注射Ad-HGF组、注射携带绿色荧光蛋白(Ad-GFP)组、注射生理盐水空白对照组,心室腔注射Ad-HGF组、注射Ad-GFP组,各组分别于第3、7、142、1、28天各处死3只检查腺病毒的分布情况及进行血管计数。结果心肌内和心室腔注射Ad-HGF组第14、21、28天可见新生血管生成数量显著增加,与对照组相比差异有统计学意义(P<0.05)。心肌内注射Ad-GFP组术后3 d心脏绿色荧光最强,14 d消失,心腔内注射Ad-GFP组术后3 d心脏可见到范围较小的微弱绿色荧光,7 d消失。结论Ad-HGF可明显促进大鼠缺血心肌微小血管生成,心肌内直接注射途径仍优于心腔内给药。
Objective To investigate the effect of adenovirus carrying Ad-HGF on the ischemic myocardium in rats. Methods 75 rats were randomly divided into three groups: Ad-HGF group, Ad-GFP group, blank control group injected with saline, Ad-HGF group, Ad-GFP group, Each group were sacrificed on the 3rd, 7th, 142th, 28th and 28th days respectively to examine the distribution of adenovirus and the blood vessel count. Results The number of neovascularization was significantly increased in Ad-HGF group (P <0.05) at the 14th, 21st and 28th days after intramyocardial and ventricular injection of Ad-HGF. The intracardiac injection of Ad-GFP group showed the strongest green fluorescence on day 3 postoperatively, and disappeared on day 14 postoperatively. The faint green fluorescence of small area was observed on the 3rd day after intracardiac injection of Ad-GFP group and disappeared on the 7th day. Conclusion Ad-HGF can significantly promote angiogenesis in ischemic myocardium of rats. The intramyocardial direct injection is still better than that of intracardiac administration.
