目的探讨当归红芪超滤物对急性心肌梗死大鼠乳酸脱氢酶(actate dehydrogenase,LDH)、乳酸脱氢酶同工酶(lactate dehydrogenase,LDH1)和热休克蛋白70(heat shock protein 70,HSP70 mRNA)表达的影响,阐明当归红芪超滤物对梗死心肌的保护作用。方法结扎左前降支建立大鼠心梗模型。将80只雄性Wistar大鼠随机分成5组,每组16只,分别为造模组(myocardial infarction,MI)、假手术组(sham-operation,SH)、低剂量组(low-dose drug,LD)、中剂量组(middle dose group,MD)、大剂量组(high-dose drug HD),各组动物分别于治疗后(36 h、14 d)断头处死,分别检测外周血LDH、LDH1和采用逆转录-聚合酶链反应(RT-PCR)技术检测大鼠心肌组织HSP70 mRNA的表达。结果与MI相比,显著降低了ST段抬高、LDH、LDH1水平(P<0.05,P<0.01)和增强了HSP70的表达(P<0.05,P<0.01)。结论当归红芪超滤物可通过抗氧化作用、降低LDH、LDH1、增加HSP70的表达发挥抗心肌缺血损伤,对大鼠心肌有明显的保护作用。
Objective To investigate the effects of Radix Angelicae Sinensis Ultrafiltration on the expression of lactate dehydrogenase (LDH), lactate dehydrogenase (LDH1) and heat shock protein 70 (HSP70) in rats with acute myocardial infarction mRNA expression, clarify the protective effect of Radix Angelicae Sinensis Ultrafiltration on myocardial infarction. Methods Left anterior descending artery was ligated to establish rat myocardial infarction model. Eighty male Wistar rats were randomly divided into 5 groups (16 rats in each group): myocardial infarction (MI), sham-operation (SH), low-dose drug Middle dose group (MD) and high-dose drug HD. The animals in each group were sacrificed at 36 h and 14 d after treatment, and the levels of LDH, LDH1, Reverse transcription polymerase chain reaction (RT-PCR) was used to detect the expression of HSP70 mRNA in rat myocardium. Results Compared with MI, ST elevation, LDH and LDH1 levels were significantly decreased (P <0.05, P <0.01) and HSP70 expression was enhanced (P <0.05, P <0.01). Conclusion Angelica Hedysari ultrafiltrated can exert anti-myocardial ischemic injury through anti-oxidation, LDH and LDH1 decrease, and increase the expression of HSP70, which has a significant protective effect on myocardium of rats.
