[目的]探讨昆仑雪菊中总黄酮粗提取物、纯化物及总多糖对人结肠癌细胞株(HCT116细胞、CACO-2细胞)增殖的抑制作用。[方法]体外培养结肠癌细胞,采用不同浓度昆仑雪菊中总黄酮粗提取物、纯化物及总多糖对其进行干预,MTT法检测细胞生长抑制率,流式细胞仪测细胞凋亡率。[结果]MTT法检测昆仑雪菊中总黄酮粗提取物、纯化物及总多糖对结肠癌细胞株的抑制作用具有浓度和时间依赖性;流式细胞仪检测昆仑雪菊总黄酮纯化物能增强结肠癌细胞株的凋亡率,且剂量与凋亡率呈正相关。[结论]昆仑雪菊总黄酮纯化物抗肿瘤活性优于总黄酮粗提物及总多糖,且对结肠癌细胞株的增殖有明显抑制作用,其作用机制可能与增强细胞凋亡有关。
[Objective] To investigate the inhibitory effect of total flavonoids, purifers and total polysaccharides from Kunlun Xueju on the proliferation of human colon cancer cell lines (HCT116 cells and CACO-2 cells). [Method] Colon cancer cells were cultured in vitro. The crude extracts of total flavonoids from Kunlun Xueju were purified and the total polysaccharides were used to intervene. MTT assay was used to detect the rate of cell growth inhibition and flow cytometry. [Result] The MTT method was used to detect the inhibitory effect of total flavonoids, purified compounds and total polysaccharides in Kunlun Xueju on the colon cancer cell lines in a concentration- and time-dependent manner. Flow cytometry was used to detect the total flavonoids in Kunlun Xueju Apoptosis rate of colon cancer cell lines, and dose and apoptosis rate was positively correlated. [Conclusion] The antitumor activity of total flavonoids from Kunlun Xueju was better than that of total flavonoids and total polysaccharides, and significantly inhibited the proliferation of colon cancer cell lines. The mechanism may be related to the enhancement of apoptosis.
