甜瓜(Cucumis melo)子叶外植体能被含双元载体的根瘤农杆菌菌株LBA4404感染。该载体内含1个NPT-Ⅱ基因(新霉素磷酸基转移酶Ⅱ基因),以供选择卡那霉素抗性的转化甜瓜细胞和1个WMV-2 CP基因(西瓜花叶病毒2号外壳蛋白基因)。卡那霉素抗性植株经PCR-Southem杂交试验表明,外源的WMV2CP基因确已通过农杆菌介导的转化途径导入甜瓜细胞。目前转基因甜瓜已开花结果。攻毒试验表明,与对照相比,转基因植株可以推迟发病时间,减轻发病程度,表现出对WMV-2具有较强的抗性。
Cucumis melo cotyledon explants can be infected with the binary vector-containing Agrobacterium tumefaciens strain LBA4404. The vector contains one NPT-II gene (neomycin phosphotransferase II gene) for selection of kanamycin-resistant transformed melon cells and one WMV-2 CP gene (Watermelon Mosaic Virus 2 Coat protein gene). Kanamycin-resistant plants by PCR-Southem hybridization test showed that exogenous WMV2CP gene indeed through Agrobacterium-mediated transformation into melon cells. The current GM melon has the flowering result. Toxicity test showed that compared with the control, the transgenic plants could delay the onset time and reduce the incidence, and showed strong resistance to WMV-2.
