目的探讨灯盏花素保护大鼠肾脏缺血-再灌注(I-R)损伤的机制。方法 SD大鼠30只,分为假手术对照(Sham)组,I-R组和灯盏花素组。免疫组织化学法检测肾脏L-选择素蛋白的表达。检测血清、肾组织一氧化氮(NO)、一氧化氮合酶(NOS)、超氧化物歧化酶(SOD)和丙二醛(MDA)水平。结果与Sham组相比,I-R组血清、肾组织MDA明显升高,SOD明显降低(P均<0.05);灯盏花素组血清、肾组织MDA低于I-R组,SOD高于I-R组(P均<0.05)。I-R组血清和肾组织NO、NOS明显高于Sham组,灯盏花素组NO、NOS水平低于I-R组(P均<0.05)。I-R组肾组织L-选择素蛋白表达明显高于Sham组,灯盏花素组L-选择素蛋白表达低于I-R组。结论灯盏花素对肾脏有保护作用。可能机制与其减少L-选择素蛋白表达,提高SOD活性,降低NOS、NO、MDA水平有关。
Objective To investigate the mechanism of breviscapine in protecting renal ischemia-reperfusion (I-R) injury in rats. Methods Thirty SD rats were divided into sham operation group, I-R group and breviscapine group. Immunohistochemistry was used to detect the expression of L-selectin protein in kidney. Serum and renal tissue were measured for levels of nitric oxide (NO), nitric oxide synthase (NOS), superoxide dismutase (SOD) and malondialdehyde (MDA). Results Compared with Sham group, MDA in serum and kidney of IR group were significantly increased (P <0.05), MDA in serum and kidney tissue of IR group were lower than those in IR group (P <0.05) <0.05). The levels of NO and NOS in serum and kidney of I-R group were significantly higher than those of Sham group, while the levels of NO and NOS in breviscapine group were lower than those in I-R group (all P <0.05). The expression of L-selectin in renal tissue of I-R group was significantly higher than that in Sham group, while the expression of L-selectin protein in breviscapine group was lower than that of I-R group. Conclusion Breviscapine has a protective effect on the kidneys. The possible mechanism is related to reducing L-selectin protein expression, increasing SOD activity and decreasing NOS, NO and MDA levels.
