目的:观察抑郁大鼠脑内细胞外信号调节激酶(ERK)通路的表达及颐脑解郁方的干预作用。方法:建立抑郁模型,采用蛋白印迹(Western Blot)实验方法,研究ERK1/2在抑郁模型大鼠脑内左前额叶皮质、海马的表达及颐脑解郁方的干预作用。结果:模型组大鼠海马、前额皮质的ERK1/2表达明显下降,与正常组相比较,有显著性差异(P<0.05,P<0.01);颐脑解郁方干预后可上调ERK1/2在脑内的表达,与模型组相比有显著性差异(P<0.05,P<0.01)。结论:抑郁模型大鼠脑内ERK1/2表达降低,颐脑解郁方可使之明显上调,颐脑解郁方的抗抑郁作用可能与上调ERK1/2信号转导通路有关。
Objective: To observe the expression of extracellular signal-regulated kinase (ERK) pathway in the brain of depression rats and the intervention effect of the Yinao Jieyu recipe. Methods: Depression model was established. The expression of ERK1 / 2 in the left prefrontal cortex and hippocampus and the effect of Yinao Jieyu prescription in depression model rats were studied by Western Blot. Results: The expression of ERK1 / 2 in hippocampus and prefrontal cortex in model group decreased significantly compared with that in normal group (P <0.05, P <0.01) There was significant difference (P <0.05, P <0.01) between the model group and the model group. Conclusion: The expression of ERK1 / 2 in the brain of depression model rats decreased, and the activity of Yinao Jieyu was significantly increased. The antidepressant effect of Yinao Jieyu Prescription may be related to the up - regulation of ERK1 / 2 signal transduction pathway.
