目的:观察白细胞介素-6(interleukin-6,IL-6)对心脏成纤维细胞(cardiacfibroblasts,CFs)增殖及p27蛋白表达的影响,探讨IL-6促CFs增殖的细胞分子生物学机制。方法:以培养的新生SD大鼠CFs为实验模型,采用四氮唑盐比色法检测细胞增殖,流式细胞分析仪技术测定细胞周期及p27蛋白表达的阳性率。结果:①1×107U/LIL-6作用48h,四氮唑盐比色法测定的CFs的A值为0.38±0.01,明显高于基础状态组(0.32±0.01),差异有显著性意义(t=-15.983,P=0.000)。②IL-6作用组CFs的S期细胞百分率(13.00±3.53)%和细胞增殖指数(PI)(29.43±1.94)%均高于基础状态组犤(7.45±0.97)%和(26.03±0.96)%犦,并有差异有显著性意义(t=-3.036,P=0.023;t=-3.142,P=0.020),而G0/G1期细胞百分率(70.58±1.94)%低于基础状态组(73.98±0.98)%,差异显著(t=3.142,P=0.020)。③IL-6作用组CFs的p27蛋白表达阳性率为(72.60±2.47)%,明显低于基础状态组(78.45±1.91)%,差异有显著性意义(t=3.753,P=0.009)。结论:IL-6能够诱导CFs增殖,其机制可能与p27蛋白表达水平下调有关。
OBJECTIVE: To observe the effects of interleukin-6 (IL-6) on the proliferation of cardiac fibroblasts (CFs) and the expression of p27 protein, and to explore the molecular mechanism of IL-6 promoting CFs proliferation. Methods: CFs from cultured neonatal SD rats were used as experimental models. Cell proliferation was measured by tetrazolium salt colorimetric assay. Cell cycle and expression of p27 protein were detected by flow cytometry. Results: ① After treated with 1 × 107 U / LIL-6 for 48 hours, the A value of CFs measured by tetrazolium salt colorimetry was 0.38 ± 0.01, which was significantly higher than that of the baseline group (0.32 ± 0.01) (t = -15.983, P = 0.000). ② The percentage of S phase cells (13.00 ± 3.53)% and cell proliferative index (PI) of CFs in IL-6 group were higher than those in basal group (7.45 ± 0.97% vs (26.03 ± 0.96)%) (T = -3.036, P = 0.023; t = -3.142, P = 0.020), while the percentage of cells in G0 / G1 phase (70.58 ± 1.94)% was lower than that in basal group (73.98 ± 0.98)%, the difference was significant (t = 3.142, P = 0.020). ③ The positive rate of p27 protein expression in CFs was (72.60 ± 2.47)% in IL-6-treated group, which was significantly lower than that in basal group (78.45 ± 1.91)%, the difference was significant (t = 3.753, P = 0.009). CONCLUSION: IL-6 can induce the proliferation of CFs, which may be related to the down-regulation of p27 protein expression.
