目的研究血必净注射液对高糖环境下大鼠腹膜间皮细胞(PMC)分泌高迁移率族蛋白1(HMGB-1)和转化生长因子β1(TGF-β1)的影响,探讨其对腹膜透析相关性腹膜纤维化的防治意义。方法胰蛋白酶消化法行PMC的原代培养和传代,经鉴定第3代细胞用于实验。随机分实验组和对照组,实验组加2.5%的葡萄糖及不同浓度的血必净,培养不同时点分别收集上清液,用ELISA法检测其中HMGB-1和TGF-β1的含量,并进行统计学分析。结果加2.5%葡萄糖的各实验组HMGB-1及TGF-β1的含量均显著高于对照组(P<0.05);加血必净的各实验组HMGB-1及TGF-β1的含量均显著低于单纯高糖组(P<0.05),且随着血必净浓度升高,HMGB-1及TGF-β1的含量下降,20mg/ml血必净组两者含量最低。结论高浓度葡萄糖可促进PMC表达HMGB-1和TGF-β1,而血必净可以拮抗高糖对PMC中HMGB-1和TGF-β1表达的上调作用,提示血必净对预防、延缓腹膜纤维化有一定意义。
Objective To study the effect of Xuebijing injection on the secretion of high mobility group box 1 (HMGB-1) and transforming growth factor β1 (TGF-β1) in rat peritoneal mesothelial cells (PMC) under high glucose conditions, and to investigate its effect on peritoneum. Dialysis-related peritoneal fibrosis prevention and control significance. Methods The primary culture and passage of PMC were performed by trypsin digestion. The third passage cells were identified and used for the experiment. Randomly divided into experimental and control groups, the experimental group added 2.5% glucose and different concentrations of Xuebijing. The culture supernatant was collected at different time points, and the contents of HMGB-1 and TGF-β1 were detected by ELISA and performed. Statistical analysis. Results The content of HMGB-1 and TGF-β1 in each experimental group plus 2.5% glucose was significantly higher than that in the control group (P<0.05). The contents of HMGB-1 and TGF-β1 in each experimental group were significantly lower. In the pure high glucose group (P<0.05), and with the increase of blood concentration, the content of HMGB-1 and TGF-β1 decreased, and the content of both of the 20 mg/ml Xuebijing group was the lowest. Conclusion High concentration of glucose promotes the expression of HMGB-1 and TGF-β1 in PMC, and Xuebijing can antagonize the up-regulation of HMGB-1 and TGF-β1 expression in PMC by high-glucose, suggesting that Xuebijing may prevent and delay peritoneal fibrosis. There is a certain significance.
