目的观察丹蒲胶囊对自身免疫性前列腺炎大鼠模型(EAPAM)炎性细胞因子的影响,探讨中药治疗慢性前列腺炎(CP)的作用机制。方法采用腹部皮下多点注射大鼠前列腺组织抗原的方法造成EAPAM,随机分成3组,每组10只,即:丹蒲胶囊组、前列泰片组、模型组,并以10只正常动物作为正常对照组。各组动物给予灌胃治疗,给药56天后处死动物,光镜观察各组动物前列腺组织的病理改变,通过酶联免疫吸附法检测各组动物血清及前列腺组织中IL-8、IL-10、TNF-α、PGE2等的水平。结果与正常对照组比较,模型组动物血清及前列腺组织中IL-8、、IL-10、TNF-α水平及前列腺组织中PGE2水平均明显升高(P<0.05~0.01)。丹蒲胶囊组血清及前列腺组织中IL-8〔(3.07±0.61)ng/L、(7.32±2.44)ng/L〕较模型组〔(4.73±1.95)ng/L、(10.14±3.64)ng/L〕降低;丹蒲胶囊组血清及前列腺组织TNF-α〔(85.34±19.20)ng/L、(87.01±15.40)ng/L〕较模型组〔(111.48±31.57)ng/L、(119.88±14.13)ng/L〕降低;丹蒲胶囊组前列腺组织中IL-10〔(34.05±7.56)ng/L〕较模型组〔(47.20±15.97)ng/L〕明显降低;丹蒲胶囊组PGE2〔(603.97±114.62)ng/L〕较模型组〔(712.58±117.10)ng/L)〕也明显降低,差异均有统计学意义(P<0.05,P<0.01)。结论丹蒲胶囊能够明显减轻模型大鼠前列腺炎症反应,调节局部免疫状态。
Objective To observe the effect of Danpu Capsule on inflammatory cytokines in the rat model of autoimmune prostatitis (EAPAM) and to explore the mechanism of traditional Chinese medicine in the treatment of chronic prostatitis (CP). Methods EAPAM was induced by intradermal multi-point injection of rat prostate tissue antigen into the abdomen and randomly divided into 3 groups, 10 in each group: Danpu capsule group, Qianlietai tablet group, model group, and 10 normal animals as normal. Control group. Animals in each group were given gavage treatment. Animals were sacrificed 56 days after administration. The pathological changes of prostate tissue in each group were observed by light microscopy. IL-8 and IL-10 in the serum and prostate tissue of each group were detected by enzyme-linked immunosorbent assay. Levels of TNF-α, PGE2, etc. Results Compared with the normal control group, the serum levels of IL-8, IL-10, TNF-α and PGE2 levels in prostate tissue were significantly higher in the model group (P<0.05-0.01). Serum and prostate tissues in Danpu capsule group had significantly higher IL-8 ((3.07±0.61) ng/L, (7.32±2.44) ng/L) than the model group ((4.73±1.95) ng/L, (10.14±3.64) ng /L〕Decrease; Danpu capsule group serum and prostate tissue TNF-α [(85.34 ± 19.20) ng / L, (87.01 ± 15.40) ng / L] than the model group [(111.48 ± 31.57) ng / L, (119.88 ±14.13) ng/L] decreased; IL-10 ((34.05±7.56) ng/L) in the prostate tissue of Danpu capsule group was significantly lower than that in the model group ((47.20±15.97) ng/L); PGE2 in Danpu capsule group. [(603.97±114.62)ng/L] was also significantly lower than that of the model group (712.58±117.10) ng/L), and the difference was statistically significant (P<0.05, P<0.01). Conclusion Danpu capsule can significantly reduce the proinflammatory reaction of model rats and regulate the local immune status.
