目的 :探讨 CD45设门在急性白血病免疫分型中的可行性和实用价值。方法 :采用CD45单抗直接免疫荧光标记和流式细胞仪对 3 2名健康人外周血白细胞及 5 6例急性白血病患者骨髓中的细胞进行了检测和分析。结果 :淋巴细胞门内 ,正常对照组相对急性白血病患者组 CD45-F ITC阳性细胞的百分比和平均荧光道数值分别为 (x± s) :(99.1± 0 .5 ) %对 (85 .2± 4.3 ) % (P<0 .0 5 )和 3 1.66±8.47对 2 .46± 1.0 7(P<0 .0 0 1) ;CD45-PE为 (99.3± 0 .7) %对 (90 .2± 6.6) % (P<0 .0 5 )和 10 6.99±2 5 .94对 13 .86± 9.5 1(P<0 .0 0 1)。急性白血病患者的原始幼稚细胞与外周血白细胞 1∶ 10 0 0混合稀释后 ,CD45/SSC参数图上 ,可清楚地对原始幼稚细胞设门。结论 :利用 CD45设门方法 ,能简单、快速、准确地识别混在外周血中的少量原始幼稚细胞 ,敏感度高达 10 -3 。
Objective: To investigate the feasibility and practical value of CD45 gating in immunological typing of acute leukemia. Methods: The bone marrow cells from 32 healthy human peripheral blood leukocytes and 56 acute leukemia patients were detected and analyzed by direct immunofluorescence labeling and flow cytometry. Results: The percentage of CD45-F ITC positive cells and the mean fluorescence channel number of lymphocytes in portal area and normal control group were (x ± s): (99.1 ± 0.5)% vs (85.2 ± 4.3% (P <0.05) and 3 1.66 ± 8.47 vs 2.46 ± 1.07 (P <0.001); CD45-PE was (99.3 ± 0.7)% vs 2 ± 6.6)% (P <0.05) and 10 6.99 ± 2 5 .94 vs 13 .86 ± 9.5 1 (P <0. 001). The original naive cells of acute leukemia patients with peripheral leukocytes 1:10 0 0 mixed dilution, CD45 / SSC parameters on the map, the original naive cells can be clearly located on the door. Conclusion: Using the CD45 portal approach, a small amount of naïve cells mixed in peripheral blood can be identified easily, rapidly and accurately with a sensitivity of 10 -3.
