目的:观察红景天苷对高原红细胞增多症大鼠的防治作用。方法:实验大鼠随机分为空白组、模型组、红景天苷高、中、低剂量组和阳性药组,测定红细胞数、血红蛋白含量、红细胞压积,全血黏度、血浆黏度,采用酶联免疫法测定一氧化氮(NO)、一氧化氮酶(NOS)、内皮素(ET)-1、血管内皮生长因子(VEGF)。结果:模型组大鼠红细胞(RBC)、血红蛋白(Hb)、血球压积(HCT)与空白组比较,显著升高(P<0.05)。与模型组比较,红景天苷高、中、低剂量具有降低上述指标的作用,尤其是中、高剂量组作用显著(P<0.05)。模型组大鼠血浆黏度、全血黏度与空白组比较,显著升高(P<0.05)。与模型组比较,红景天苷高、中、低剂量具有降低全血黏度、血浆黏度的作用,尤其是高剂量作用显著(P<0.05)。模型组大鼠NO、NOS与空白组比较显著降低(P<0.05),ET-1、VEGF显著升高(P<0.05)。与模型组比较,红景天苷高剂量具有显著升高NO、NOS,降低VEGF的作用(P<0.05);红景天苷高、中剂量具有显著降低ET-1的作用(P<0.05)。结论:红景天苷对高原红细胞增多症大鼠有较好的防治作用。
Objective: To observe the effect of salidroside on rats with altitude polycythemia. Methods: The rats were randomly divided into blank group, model group, salidroside high, medium and low dose groups and positive drug group. The number of red blood cells, hemoglobin, hematocrit, whole blood viscosity and plasma viscosity were measured. The levels of nitric oxide (NO), nitric oxide synthase (NOS), endothelin (ET) -1 and vascular endothelial growth factor (VEGF) were measured by immunoassay. Results: Compared with blank group, RBC, Hb and HCT in model group were significantly increased (P <0.05). Compared with the model group, salidroside high, medium and low doses have the effect of reducing the above indicators, especially in the medium and high dose groups (P <0.05). Compared with the blank group, the viscosity and whole blood viscosity of the model group were significantly increased (P <0.05). Compared with model group, salidroside had the effect of lowering whole blood viscosity and plasma viscosity, especially high dose (P <0.05). NO and NOS in model group were significantly lower than those in blank group (P <0.05), while ET-1 and VEGF were significantly increased (P <0.05). Compared with the model group, salidroside at high dose significantly increased NO, NOS, decreased the role of VEGF (P <0.05); Salidroside high and medium dose significantly reduced the role of ET-1 (P <0.05) . Conclusion: Salidroside has good preventive and therapeutic effects on rats with altitude polycythemia.
