目的:研究ECRG4基因在胃腺癌组织、癌旁组织及正常胃组织中的表达差异及其与患者临床病理特征的相关性。方法:收集手术切除并经病理证实的胃腺癌组织石蜡标本58例作为胃癌组,癌旁组织及正常胃组织各15例作为癌旁组、正常组,用免疫组织化学SP法检测胃腺癌组织与癌旁及正常组织中ECRG4编码蛋白的表达情况,用SPSS19.0统计软件对ECRG4蛋白的表达差异及其与患者的临床病理特征进行统计学分析。结果:ECRG4蛋白在胃癌组中阴性表达29例、弱阳性表达29例、未见阳性及强阳性表达;癌旁组织中阴性表达0例、弱阳性表达5例、阳性表达7例、强阳性表达3例;正常胃组织中阴性表达1例、弱阳性表达4例、阳性表达2例、强阳性表达8例。三者E-CRG4蛋白表达差异具有统计学意义(P<0.001)。两两比较后认为正常胃组织与胃癌组织、胃癌组织与癌旁组织E-CRG4蛋白表达差异具有统计学意义(P<0.05),而正常胃组织与胃癌癌旁组织ECRG4蛋白表达差异不具有统计学意义(P>0.05)。ECRG4的表达与胃癌患者的分化程度有关,分化程度越低者其表达缺失率越高(P<0.05);ECRG4蛋白表达与病人年龄、性别、肿瘤发生部位、TNM分期及是否淋巴结转移无关(P>0.05)。结论:ECRG4蛋白在胃腺癌组织中表达明显下调或缺失,而在胃正常组织中普遍表达。ECRG4蛋白表达与患者肿瘤的分化程度有关,分化程度越低者其表达缺失率越高。
Objective: To investigate the difference of ECRG4 expression in gastric adenocarcinoma, paracarcinoma and normal gastric tissue and its correlation with clinicopathological features. Methods: Fifty-eight cases of gastric adenocarcinoma and paraffin-embedded specimens of gastric adenocarcinoma were collected from 15 patients with gastric cancer, adjacent normal tissue and 15 normal gastric tissues respectively. The immunohistochemical SP method was used to detect the expression of The expression of ECRG4 protein in paracancerous tissues and normal tissues was detected by SPSS19.0 software. The differences of ECRG4 protein expression between the two groups were statistically analyzed with the clinicopathological features. Results: The negative expression of ECRG4 protein was found in 29 cases of gastric cancer group and 29 cases of weakly positive expression, but no positive or strong positive expression was found in the gastric cancer group. There were 0 negative expression in paracancerous tissues, 5 weakly positive expression and 7 positive expression. 3 cases; normal gastric tissue negative expression in 1 case, weakly positive expression in 4 cases, positive expression in 2 cases, strong positive expression in 8 cases. The difference of E-CRG4 protein expression was statistically significant (P <0.001). The comparison of the expression of E-CRG4 protein between normal gastric tissues and gastric cancer tissues, gastric cancer tissues and adjacent normal tissues was statistically significant (P <0.05), while the difference of ECRG4 protein expression between normal gastric tissues and adjacent non-cancerous tissues was not statistically significant Significance (P> 0.05). The expression of ECRG4 was correlated with the degree of differentiation in patients with gastric cancer. The lower the degree of differentiation was, the lower the expression of ECRG4 was (P <0.05). The expression of ECRG4 was not associated with age, gender, tumor location, TNM stage and lymph node metastasis > 0.05). Conclusion: The expression of ECRG4 protein in gastric adenocarcinoma was significantly down-regulated or down-regulated, while it was generally expressed in normal gastric tissues. ECRG4 protein expression in patients with the degree of tumor differentiation, the lower the degree of differentiation, the higher the rate of loss of expression.
