目的 建立病毒性心肌炎细胞感染模型。 方法 以柯萨奇 B3m病毒感染新生 SD大鼠的培养心肌细胞 ,观察感染后细胞病变、搏动情况及超微结构的变化 ,检测培养液中心肌酶的改变 ,测定 DNA含量。 结果 培养的 SD大鼠搏动心肌细胞感染柯萨奇 B3m病毒后 ,逐步出现细胞病变 ,细胞停止搏动 ,电镜下可见细胞线粒体肿胀 ,肌原纤维紊乱 ,Z线模糊 ,髓质体增多 ,细胞周期分析显示细胞受阻于 G1 期。 结论 该模型可作为体外研究病毒性心肌炎的基础。
Objective To establish a viral myocarditis cell infection model. Methods Coxsackie B3m virus was used to infect neonatal SD rats to observe the changes of cellular pathological changes, pulsatility and ultrastructure of infected SD rats. The changes of myocardial enzymes in the culture fluid were detected to determine the DNA content. Results Cortical cardiomyocytes of SD rats were infected with Coxsackie B3m virus, and gradually developed cytopathic effect. The cells stopped beating. The mitochondria swelling, myofibril disorganization, fuzzy Z line, increased medulla, cell cycle analysis Showing cells blocked in G1 phase. Conclusion This model can be used as a basis for studying viral myocarditis in vitro.
