麝香为雄麝香腺囊的分泌物,其水溶性提取物musclide具有强心作用,并能激活蛋白激酶。作者发现其强心作用是由于它强化β-受体。作者曾提出musclide的强心作用机制是由于二酰基甘油作用和forskolin样作用。麝香中有香味的水不溶成分无强心作用。最近作者从麝香分得一个强蛋白激酶激活剂musclide-Al,化学结构为6-甲基-2,5-庚二醇-5-亚硫酸盐及其有关化合物。之后又合成了其同系物,并比较了这些化合物的强心作用。Musclide-Al(30μg/ml,130μmol)较musclide和麝香的强心作用为强。若预先服蛋白激酶抑制剂,musclide的强心作用则被抑制。
Musk is the secretion of the male fragrant gland capsule, and its water-soluble extract musclide has a cardiotonic effect and can activate protein kinases. The authors found that its cardiotonic effect was due to its potentiation of β-receptors. The authors have suggested that the cardiac mechanism of musclide is due to diacylglycerol effects and forskolin-like effects. The aromatic water-insoluble ingredients in musk have no heart effect. Recently, the authors isolated musclide-Al, a strong protein kinase activator from Musk, with the chemical structure of 6-methyl-2,5-heptanediol-5-sulphite and related compounds. Afterwards, their homologues were synthesized and the cardiotonic effects of these compounds were compared. Musclide-Al (30 μg/ml, 130 μmol) was stronger than Musclide and Musk. If the protein kinase inhibitor is pre-administered, the cardiac action of musclide is inhibited.
