目的研究肿瘤转移相关基因1(MTA-1)与肿瘤转移抑制基因(KISS-1)表达蛋白在膀胱癌中的表达及临床病理意义。方法采用免疫组化SP法检测65例膀胱癌组织和15例膀胱正常组织中MTA-1和KISS-1的表达,结合临床资料及组织病理学特点探讨MTA-1和KISS-1基因表达与膀胱癌浸润和转移的关系。结果 MTA-1蛋白在膀胱癌组织阳性表达率明显高于膀胱正常组织(53.85%vs.6.67%)(P<0.01);KISS-1蛋白在膀胱癌组织阳性表达率明显低于膀胱正常组织(40.00%vs.86.67%)(P<0.01);其基因表达与病理分级、临床分期、淋巴结转移密切相关(P<0.05)。膀胱癌组织中MTA-1与KISS-1蛋白表达呈负相关(P<0.01)。结论 MTA-1蛋白高表达和KISS-1蛋白低表达与膀胱癌的浸润、转移密切相关,可作为诊断膀胱癌患者转移、复发和判断预后的重要参考指标,有望成为膀胱癌基因治疗的新靶点。
Objective To investigate the expression and clinicopathological significance of MTA-1 and KISS-1 in bladder cancer. Methods Immunohistochemical SP method was used to detect the expression of MTA-1 and KISS-1 in 65 cases of bladder cancer tissues and 15 cases of normal bladder tissues. The expressions of MTA-1 and KISS-1 gene and the expression of KISS-1 were detected by clinical data and histopathological features. Invasion and metastasis of cancer. Results The positive expression rate of MTA-1 protein in bladder cancer tissues was significantly higher than that in normal bladder tissues (53.85% vs.6.67%, P <0.01). The positive expression rate of KISS-1 protein in bladder cancer tissues was significantly lower than that in normal bladder tissues 40.00% vs.86.67%) (P <0.01). The gene expression was closely related to pathological grade, clinical stage and lymph node metastasis (P <0.05). The expression of MTA-1 and KISS-1 in bladder cancer tissues was negatively correlated (P <0.01). Conclusion The high expression of MTA-1 protein and low expression of KISS-1 protein are closely related to the invasion and metastasis of bladder cancer, which may serve as an important reference for diagnosis of metastasis, recurrence and prognosis in patients with bladder cancer and may be a new target for gene therapy of bladder cancer point.
