目的探讨白细胞介素-17(IL-17)A~F(IL-17A、IL-17B、IL-17C、IL-17D、IL-17E、IL-17F)在慢性乙型病毒性肝炎(乙肝)及肝硬化中的表达情况及临床意义。方法收集66例慢性乙肝患者(慢性乙肝组)和50例乙肝肝硬化(肝硬化组)患者的血清及粪便,以及30例健康志愿者(对照组)的血清。用荧光定量PCR检测乙肝病毒(HBV)DNA,用酶联免疫吸附试验法(ELISA)检测IL-17A~F、IL-1β、IL-6、趋化因子-13。比较3组IL-17A~F的表达情况及与细胞因子、肝功能、HBV DNA的相关性。结果慢性乙肝组及肝硬化组的IL-17A~F均明显高于对照组(P<0.01),但慢性乙肝组与肝硬化组间差异无统计学意义(P>0.05)。慢性乙肝组IL-17A~F与IL-1β、IL-6、CXCL-13呈正相关,与总胆红素呈负相关;IL-17C与总胆汁酸呈正相关;IL-17B,IL-17D,IL-17E与粪便HBV DNA呈正相关。肝硬化组IL-17B与IL-1β呈正相关;IL-17C与谷草转氨酶、碱性磷酸酶呈正相关,IL-17D与碱性磷酸酶呈正相关;IL-17A~F与血清及粪便HBV DNA均无相关性(P>0.05)。结论慢性乙肝及肝硬化患者均存在IL-17A~F高表达,但临床意义不完全相同。
Objective To investigate the effect of interleukin-17 (IL-17) A to F on chronic hepatitis B (hepatitis B) And cirrhosis of the expression and clinical significance. Methods The sera from 66 patients with chronic hepatitis B (chronic hepatitis B) and 50 patients with cirrhosis (cirrhosis) were collected and serum from 30 healthy volunteers (control group) were collected. The HBV DNA was detected by real-time quantitative PCR and IL-17A-F, IL-1β, IL-6 and chemokine-13 were detected by enzyme-linked immunosorbent assay (ELISA) The expression of IL-17A-F and the correlation with cytokines, liver function and HBV DNA in 3 groups were compared. Results The levels of IL-17A ~ F in chronic hepatitis B group and cirrhosis group were significantly higher than those in control group (P <0.01), but there was no significant difference between chronic hepatitis B group and cirrhosis group (P> 0.05). There was a positive correlation between IL-17A-F and IL-1β, IL-6 and CXCL-13 in chronic hepatitis B group and a negative correlation with total bilirubin; IL-17C was positively correlated with total bile acid; IL- IL-17E and stool HBV DNA was positively correlated. IL-17B was positively correlated with IL-1β in cirrhosis group; IL-17C was positively correlated with aspartate aminotransferase and alkaline phosphatase; IL-17D was positively correlated with alkaline phosphatase; IL- No correlation (P> 0.05). Conclusion The expression of IL-17A ~ F is high in patients with chronic hepatitis B and cirrhosis, but the clinical significance is not exactly the same.
