用体外微量测定法比较了青蒿素、双氢青蒿素、蒿甲醚和咯萘啶对4株恶性疟原虫的敏感性,其中F32株(坦桑尼亚)、7G8株(巴西)、洪都拉斯株(洪都拉斯)为氯喹敏感株,D7株(坦桑尼亚)为氯喹抗性株。培养基采用WHO提供的特殊RPMI培养基(含低浓度对氨基苯甲酸和叶酸),另加Hepes缓冲液、NaHCO_3、人O型血清、谷酰胺和正常O型红细胞。试验前先将待测药品按比例稀释,终浓度为10~(-4)~10~(-9)mol/L,以100μl/井加入微量测定板,置37℃培养箱干燥24h,再
The sensitivity of four strains of Plasmodium falciparum to artemisinin, dihydroartemisinin, artemether and pyronaridine was compared with in vitro microassay, of which F32 (Tanzania), 7G8 (Brazil), Honduras ( Honduras) as chloroquine sensitive strains, D7 strain (Tanzania) as chloroquine resistant strains. The medium used was the special RPMI medium (containing low concentrations of para-aminobenzoic acid and folic acid) provided by the WHO plus Hepes buffer, NaHCO 3, human O-type serum, glutamine and normal O-type erythrocytes. Before the test, the drugs to be tested should be diluted proportionally to a final concentration of 10 ~ (-4) ~ 10 ~ (-9) mol / L. Add 100μl / well to the micro assay plate and set in a 37 ℃ incubator for 24 hours.
