目的建立高效液相色谱法测定肉制品中诱惑红的方法。方法样品经石油醚去除油脂,乙醇-氨-水超声提取诱惑红,亚铁氰化钾、乙酸锌沉淀蛋白,再经聚酰胺吸附提取诱惑红制成水溶液,注入高效液相色谱经紫外检测器,检测诱惑红含量。结果本法线性范围0.002~0.1mg/ml,检出限1mg/kg,RSD3.25%~4.66%;回收率95.6%~98.8%,精密度和准确度好。结论采用石油醚去除油脂,乙醇-氨-水超声提取诱惑红,亚铁氰化钾、乙酸锌沉淀蛋白,再经聚酰胺吸附提取诱惑红。可消除脂肪、蛋白质与聚酰胺粉竞争吸附诱惑红色素,提高样品中诱惑红色素的检出量。通过加入低、中、高不同含量的诱惑红色素,测定回收率95.6%~98.8%。本方法可以用于测定肉制品中诱惑红含量。
Objective To establish a method for the determination of temptation red in meat products by high performance liquid chromatography. Methods Samples were extracted with petroleum ether, and the templated red, potassium ferrocyanide and zinc acetate were precipitated by ethanol-ammonia-water ultrasonic extraction, and then the aqueous solution was made by polyamide adsorption and extraction temptation. The samples were injected into high performance liquid chromatography , Test the temptation to red content. Results The linear range of this method was 0.002 ~ 0.1mg / ml with the detection limit of 1mg / kg and RSD of 3.25% ~ 4.66%. The recoveries ranged from 95.6% to 98.8%. The precision and accuracy were good. Conclusion Petroleum ether was used to remove oil and fat, and ethanol, ammonia and water were used to extract temptation red, potassium ferrocyanide and zinc acetate precipitated protein, and then the temptation red was extracted by polyamide adsorption. Elimination of fat, protein and polyamide powder competition adsorption temptation of red pigment, to increase the detection of seductive red pigment in the sample. By adding low, medium and high content of different temptation of red pigment, the determination of recovery of 95.6% to 98.8%. The method can be used to determine the seduction red content of meat products.
