目的研究灰树花多糖(GFP)抗实验性肉瘤作用的效果及其对肿瘤坏死因子(TNF-α)、白细胞介素-2(IL-2)的影响,探讨其作用机制。方法将瘤细胞悬液接种于小鼠右前肢腋皮下制备荷瘤小鼠动物模型,分为GFP高、中、低剂量组和环磷酰胺(Cy)组、生理盐水组。以高、中、低剂量GFP溶液和环磷酰胺分别灌胃10d,检测小鼠肿瘤的抑制率,小鼠血清中TNF-α、IL-2的水平变化。结果3种剂量的GFP、环磷酰胺对S180荷瘤小鼠的抑瘤率分别为34.37%、42.67%、47.92%和41.5%,GFP组荷瘤小鼠血清内TNF-α、IL-2含量明显高于Cy组和生理盐水组(P<0.05)。结论GFP具有抗肿瘤作用,TNF-α、IL-2的释放可能是其抗肿瘤作用的机制之一。
Objective To study the effect of Grifola frondosa polysaccharide (GFP) on experimental sarcoma and its effect on tumor necrosis factor (TNF-α) and interleukin-2 (IL-2), and to explore its mechanism of action. Methods A tumor-bearing mouse model was prepared by inoculating tumor cell suspension into mouse right forelimb subcutaneously and divided into GFP high, medium and low dose groups, cyclophosphamide (Cy) group and physiological saline group. High-, medium-, low-dose GFP solution and cyclophosphamide were intragastrically administered for 10 days. The inhibitory rate of tumor in mice and the levels of TNF-α and IL-2 in mice serum were detected. Results The tumor inhibition rates of 3 doses of GFP and cyclophosphamide on S180 tumor-bearing mice were 34.37%, 42.67%, 47.92% and 41.5%, respectively. The levels of serum TNF-α and IL-2 in GFP-bearing mice It was significantly higher than Cy group and saline group (P<0.05). Conclusion GFP has anti-tumor effect. The release of TNF-α and IL-2 may be one of the mechanisms of its anti-tumor effect.
