用ABC免疫组织化学法观察乳酸脱氢酶C_4(LDH-C_4)在性成熟后的Wistar大鼠生精上皮细胞中的定位。结果表明:LDH-C_4定位于粗线期初级精母细胞以后的各级生精细胞中,精原细胞中缺如。显微分光光度分析表明,随着精子的发生,LDH-C_4的相对含量逐渐增加,精母细胞中为0.55±0.09,精子细胞中为0.86±0.13,精子中为0.94±0.04,三者之间有显著性差异(P<0.01)。ABC免疫电镜显示LDH-C_4在大鼠精子的细胞质、线粒体及部分细胞膜上均有分布。免疫荧光显示LDH-G_4在精子表面主要定位于尾部的主段和末段,少量分布于精子头冠部。作者对LDH-C_4细胞定位的生理意义作了讨论。
The localization of lactate dehydrogenase C_4 (LDH-C_4) in the spermatogenic epithelial cells of sexually mature Wistar rats was observed by ABC immunohistochemistry. The results showed that LDH-C_4 localized in spermatogenic cells at all stages after primary spermatocytes in pachytene stage, but not in spermatogonia. Micro-spectrophotometry analysis showed that the relative content of LDH-C_4 gradually increased with the development of spermatozoa, which was 0.55 ± 0.09 in spermatocytes, 0.86 ± 0.13 in spermatids and 0.94 ± 0.04 in sperms There was significant difference (P <0.01). ABC immunoelectron microscopy showed that LDH-C_4 was distributed in the cytoplasm, mitochondria and some cell membranes in rat sperm. Immunofluorescence showed that LDH-G_4 mainly localized in the main segment and the terminal segment of the tail on the sperm surface, with a small amount distributed in the sperm head crown. The authors discuss the physiological significance of LDH-C_4 cell localization.
