本文利用体外培养血管内皮细胞的方法,观察了酵母多糖活化的多形核白细胞(PMN)对内皮细胞的影响。本研究发现:将活化的PMN与内皮细胞孵育,10h内皮细胞出现核固缩等变化,培养液中LDH升高;24h内皮细胞变化更为明显,细胞变性坏死,培养液中LDH和6-酮-PGF_1α含量均明显升高。予先向培养液中分别加入超氧化物歧化酶(SOD)或过氧化氢酶(CAT),内皮细胞仍有损害,将SOD和CAT予先联合加入,可保护内皮细胞免受PMN的损害。据此认为:活化PMN引起的内皮细胞损伤可能主要系羟自由基(OH·)所致。
In this study, the effects of zymosan activated polymorphonuclear leukocytes (PMN) on endothelial cells were observed by in vitro culture of vascular endothelial cells. This study found: activated PMN and endothelial cells incubated 10h endothelial cells nuclear pyknosis and other changes in the culture medium LDH increased; 24h endothelial cell changes more pronounced cell degeneration and necrosis, LDH and 6-ketone in the culture medium -PGF_1α levels were significantly increased. Preaddition to the culture medium were added superoxide dismutase (SOD) or catalase (CAT), endothelial cells are still damaged, SOD and CAT pre-combined to protect endothelial cells from PMN damage. Therefore, it is concluded that endothelial cell injury induced by activated PMN may be mainly caused by hydroxyl radical (OH ·).
