目的: 探讨 bcl2 基因对人角膜上皮细胞生存过程的影响。方法: 用反转录聚合酶链式反应检测bcl2 m R N A 在人角膜上皮细胞中的表达情况。用荧光显微镜、电镜、流式细胞仪观察 bcl2 单克隆抗体对细胞凋亡发生的影响。结果: 用反转录聚合酶链式反应获得 332bp 的扩增带。荧光显微镜观察, 十个随机视野, 实验组与对照组凋亡细胞比例分别为 312% 和179% (u 检验, P< 001)。流式细胞仪检测的结果接近光镜下的结果, 凋亡细胞比例分别为 324% 和 198% 。电镜下可见典型的凋亡细胞。结论:人角膜上皮细胞有 bcl2 m R N A 的表达, bcl2 基因产物具有阻止人角膜上皮细胞凋亡发生的作用。
Objective: To investigate the effect of bcl2 gene on the survival of human corneal epithelial cells. Methods: Reverse transcription-polymerase chain reaction was used to detect the expression of bcl-2 mRNA in human corneal epithelial cells. Fluorescence microscopy, electron microscopy, flow cytometry bcl 2 monoclonal antibody on apoptosis. Results: A 332 bp amplification band was obtained by reverse transcription-polymerase chain reaction. Fluorescence microscopy, ten random fields of vision, the experimental group and the control group apoptotic cells were 31 2% and 17 9% (u test, P <0 01). Flow cytometry results close to the light microscope results, the proportion of apoptotic cells were 32 4% and 19 8%. A typical apoptotic cells can be seen under electron microscope. Conclusion: Human corneal epithelial cells have bcl 2 m R N A expression, bcl 2 gene products have a role in preventing human corneal epithelial cells from apoptosis.
