目的:研究β-连环蛋白(β-cat)、细胞周期蛋白D1(cyclin D1)和c-myc基因(c-myc)在正常宫颈组织、宫颈上皮内瘤变(CIN)及宫颈浸润性鳞癌中的表达是否与宫颈癌发生及发展有关。方法:采用PV-9000二步法免疫组化检测试剂,检测正常宫颈组织、CIN和宫颈浸润性鳞癌组织中β-cat、cyclin D1和c-myc的表达。结果:β-cat在正常宫颈组织胞膜中的表达呈强阳性,而在CIN和宫颈浸润性鳞癌胞膜中的表达明显减低,表现为胞质或胞核的异常表达增强,同时cyclin D1和c-myc在CIN和宫颈浸润性鳞癌中表达增强,且与CIN和宫颈浸润性鳞癌发生发展有关。三者在正常宫颈组织、CIN和宫颈浸润性鳞癌中的表达率[分别为4%(2/50)、40%(20/50)、76%(38/50);2%(1/50)、32%(16/50)、58%(29/50)和8%(4/50)、40%(20/50)、68%(34/50)]差异有统计学意义(P均<0.01)。β-cat胞质胞核的异常表达与cyclin D1和c-myc的高表达在CIN和宫颈浸润性鳞癌中均呈显著正相关(相关系数r=0.999)。结论:β-cat的异常表达参与宫颈浸润性鳞癌的发生,并有可能通过激活cyclin D1和c-myc引起细胞增殖和分化失控,促使肿瘤发生、发展。在宫颈癌的发生、发展机制中可能起重要作用。三者同时检查可以作为预测宫颈癌的发生、发展的分子生物学指标。

The abnormal expression of β-cat cytoplasmic nucleus and cyclin D1 and c-myc overexpression in CIN and invasive cervical squamous cell carcinoma showed a significant positive correlation (correlation coefficient r = 0.999). CONCLUSION: Abnormal expression of β-cat is involved in the development of invasive cervical squamous cell carcinoma and it is possible that cyclin D1 and c-myc may induce uncontrolled cell proliferation and differentiation and promote tumorigenesis and progression. In cervical cancer occurrence and development mechanism may play an important role. Three simultaneous examination can be used as a prediction of the occurrence and development of cervical cancer molecular biological indicators.
