目的探讨内皮素受体拮抗剂BQ-123对肺纤维化大鼠Ⅰ型和Ⅲ型胶原表达的影响。方法实验分为模型组、治疗组及对照组三组。模型组用博莱霉素5mg/kg气管内注射复制大鼠肺纤维化模型,治疗组予以内皮素受体拮抗剂BQ-12350μg/kg每周2次腹腔注射,对照组以等量生理盐水替代。造模后第28d处死大鼠,用半定量RT-PCR测定Ⅰ型和Ⅲ型胶原信使核糖核酸(mRNA)的表达,免疫组化法测定Ⅰ型和Ⅲ型胶原蛋白的表达。结果模型组Ⅰ型和Ⅲ型胶原mRNA表达均显著高于对照组(P<0.01),治疗组Ⅰ型和Ⅲ型胶原表达均有所下降,较之模型组有统计学差异(P<0.05)。免疫组化提示模型组Ⅰ型和Ⅲ型胶原蛋白表达增加,治疗组较之模型组有不同程度改善。结论肺纤维化大鼠Ⅰ型和Ⅲ型胶原表达增加,应用内皮素受体拮抗剂治疗可抑制上述胶原的表达。
Objective To investigate the effect of endothelin receptor antagonist BQ-123 on the expression of type I and type III collagen in rats with pulmonary fibrosis. Methods The experiment was divided into three groups: model group, treatment group and control group. Model group with bleomycin 5mg / kg intratracheal injection of rat pulmonary fibrosis model, the treatment group to endothelin receptor antagonist BQ-12350μg / kg twice a week by intraperitoneal injection, the control group with an equal volume of saline instead . Rats were sacrificed on the 28th day after the model was established. The mRNA expression of type I and type III collagen messenger RNA was detected by semi-quantitative RT-PCR. The expression of type I and type III collagen was determined by immunohistochemistry. Results The mRNA expression of type I and type III collagen in the model group was significantly higher than that in the control group (P <0.01). The expressions of type I and type III collagen in the treatment group were significantly lower than those in the model group (P <0.05) . Immunohistochemistry showed that the expression of type I and type III collagen in the model group was increased, and the treatment group was improved to some extent compared with the model group. Conclusion The expression of type Ⅰ and type Ⅲ collagen in pulmonary fibrosis rats is increased. The treatment with endothelin receptor antagonist can inhibit the expression of collagen above.
