目的:研究RFP(Ret Finger Protein,RFP)在人、鼠正常组织及食管鳞状细胞癌组织中的分布。方法:本研究运用 实时定量PCR方法并以18SrRNA作为内对照分析了RFP在人子宫颈、子宫内膜、睾丸、胃、食管和脑组织以及在鼠肺、肾、子 宫、卵巢、胸腺、心、肌肉、睾丸、皮肤、食管、脾、肝和肠等正常组织和人食管鳞状细胞癌组织中的表达。结果:RFP蛋白只在人 和鼠的睾丸组织中高表达,而在其他正常组织中低表达,在20例人食管鳞状细胞癌组织标本中检测到13例RFP表达上调, 其中9例有显著意义(P<0.05)。结论:由于RFP蛋白在正常组织中只高表达于睾丸组织,而约65%人食管鳞状细胞癌组织 中该蛋白的表达上调。上述结果提示RFP蛋白可能是治疗人食管鳞状细胞癌的一个潜在的分子靶点。
Objective: To study the distribution of RFP (Ret Finger Protein) in human and mouse normal tissues and esophageal squamous cell carcinoma tissues. Methods: In this study, real-time quantitative PCR method and 18SrRNA as an internal control of RFP in human cervix, endometrium, testis, stomach, esophagus and brain tissue and lung, kidney, uterus, ovary, thymus, Muscle, testis, skin, esophagus, spleen, liver and intestine and other normal tissues and human esophageal squamous cell carcinoma. Results: RFP protein was highly expressed in human and mouse testis tissue, but low in other normal tissues. RFP expression was detected in 13 of 20 human esophageal squamous cell carcinoma specimens, of which 9 were significant (P <0.05). CONCLUSIONS: RFP is highly expressed in testis tissue in normal tissues and is up-regulated in about 65% of human esophageal squamous cell carcinomas. The above results suggest that RFP protein may be a potential molecular target for the treatment of human esophageal squamous cell carcinoma.
