目的:研究正常人骨髓成纤维样基质细胞HFCL对 急性髓细胞白血病HL 60细胞cxcr4基因表达的影响及意义. 方法:采用流式细胞仪检测细胞周期,NBT还原实验和 CD11b,CD14,CD13,CD33细胞表面抗原的测定作为细胞分 化的指标;采用半定量RT PCR,Northernblot检测cxcr4基因 的表达.结果:HL 60细胞与HFCL细胞共培养后,NBT阳性 细胞增高,CD11b和CD14的表达增多,并有明显的统计学意 义;而且cxcr4mRNA的表达下调.结论:HFCL细胞能诱导 HL 60细胞部分向单核细胞分化,并使趋化因子受体cxcr4表 达下调.
Objective: To study the effect and significance of normal human bone marrow fibroblastoid stromal cells (HFCL) on cxcr4 gene expression in acute myeloid leukemia HL-60 cells.Methods: Flow cytometry was used to detect cell cycle, NBT reduction and CD11b, CD14, CD13, CD33 Cell surface antigen was used as an indicator of cell differentiation.Expression of cxcr4 gene was detected by semi-quantitative RT-PCR and Northern blotting.Results: After co-cultured with HL-60 cells, the number of NBT positive cells increased and the expression of CD11b and CD14 increased And the expression of cxcr4mRNA was down-regulated.Conclusion: HFCL cells can induce partial differentiation of HL60 cells into monocytes and down-regulate the expression of chemokine receptor cxcr4.
