目的优化楮桃叶配方颗粒提取工艺,建立楮桃叶配方颗粒质量标准。方法采用L9(34)正交设计对提取工艺进行优化,TLC法进行定性鉴别,HPLC法对牡荆素进行含量测定。结果楮桃叶配方颗粒的提取工艺为加水量10倍,煎煮次数2次,煎煮时间1h,浸膏得率为18.29%,牡荆素提取率可达68.13%;薄层色谱中可检出特征斑点;牡荆素进样量在0.0128~0.2052μg范围内线性关系良好(r=1.0000),平均回收率为100.32%,RSD为0.88%,n=6。结论提取工艺科学合理,TLC法及HPLC法易于操作,重复性好,可有效控制楮桃叶配方颗粒的质量。
OBJECTIVE To optimize the extraction technology of recipe peach leaves and establish the quality standard of recipe peach leaves. Methods The orthogonal design of L9 (34) was used to optimize the extraction process. TLC was used for qualitative identification. The content of vitexin was determined by HPLC. Results The extraction of peach leaf formula particles was 10 times of water addition, 2 times of boiling, 1 hour of boiling and 18.29% of extraction. The extraction rate of vitexin was 68.13% (R = 1.0000), the average recovery was 100.32%, RSD was 0.88%, n = 6. Conclusion The extraction process is scientific and reasonable, TLC method and HPLC method are easy to operate with good repeatability, which can effectively control the quality of Apium petiole leaf formula granules.
