目的考察半枝莲总黄酮对巨噬细胞炎症模型中MAPK信号通路的影响。方法半枝莲醇提物经乙醇提取,乙酸乙酯萃取及大孔树脂梯度洗脱后获得半枝莲总黄酮。LPS刺激巨噬细胞构建炎症模型。Western Blot检测药物对MAPK信号通路的影响。实时定量PCR法及ELISA法分别检测细胞因子的基因表达水平及含量的变化。MTT法检测药物对细胞生长的影响。结果半枝莲总黄酮能够抑制MAPK信号通路中JNK/SAPK的磷酸化水平,并能降低细胞因子IL-6、IL-1β的含量。结论初步明确半枝莲总黄酮的抗炎作用机制。
Objective To investigate the effect of Scutellaria barbata flavonoids on MAPK signal pathway in macrophage inflammatory model. Methods Scutellaria barbata extract was ethanol extract, ethyl acetate extract and gradient elution macroporous resin obtained Scutellaria barbata total flavonoids. LPS stimulates macrophages to construct an inflammatory model. Western Blot detection of drugs on the MAPK signaling pathway. Real-time quantitative PCR and ELISA were used to detect the gene expression level and content of cytokines. The effects of drugs on cell growth were detected by MTT assay. Results Scutellaria barbata flavonoids could inhibit the phosphorylation of JNK / SAPK in MAPK signaling pathway and decrease the levels of cytokines IL-6 and IL-1β. Conclusion The anti-inflammatory mechanism of total flavonoids from Scutellaria barbata L. was initially clarified.
