目的 了解Ⅰ型与Ⅱ型 5α 还原酶在良性增生前列腺组织中的表达情况 ,为前列腺增生的药物治疗提供理论依据。方法 用免疫组织化学及RT PCR方法对 15例正常前列腺组织和 2 5例良性增生前列腺组织中的两型 5α 还原酶同工酶的表达进行定性及半定量分析。结果 良性前列腺增生组织及正常前列腺组织中均存在两型 5α 还原酶的表达 ,阳性染色主要位于腺上皮细胞的胞质内。增生前列腺组织中Ⅱ型 5α 还原酶mRNA的表达水平明显高于正常前列腺组织 (分别为 1 5 0±0 4 2及 0 98± 0 32 ,P <0 0 1) ,而Ⅰ型还原酶的表达相对较弱且与正常前列腺组织比较差异无显著意义 (分别为 0 6 2± 0 31与 0 5 1± 0 2 4P >0 0 5 )。Ⅱ型 5α 还原酶在增生前列腺组织中的表达水平与腺体的体积及腺体所在的部位相关。结论 Ⅱ型 5α 还原酶在良性前列腺增生的发生、发展过程中起着重要的作用 ,而Ⅰ型还原酶在良性增生组织中表达的意义需要进一步研究。
Objective To understand the expression of type Ⅰ and type Ⅱ 5α reductase in benign prostatic hyperplasia and to provide a theoretical basis for the drug treatment of benign prostatic hyperplasia. Methods The expression of two types of 5α-reductase isozymes in 15 normal prostate tissues and 25 benign prostatic hyperplasia tissues were detected by immunohistochemistry and RT-PCR. Results Both benign prostatic hyperplasia and normal prostatic tissues showed the expression of two types of 5α-reductase. The positive staining was mainly located in the cytoplasm of glandular epithelial cells. The expression of type II 5α-reductase mRNA in proliferative prostate was significantly higher than that in normal prostate tissue (150 ± 0 4 2 and 0 98 ± 0 32, respectively, P 0 01), while the expression of type Ⅰ reductase Relatively weak and no significant difference compared with normal prostate tissue (0 6 2 ± 0 31 and 0 5 1 ± 0 2 4 P> 0 0 5, respectively). The expression level of type II 5α-reductase in hyperplastic prostatic tissues correlates with the size of the gland and the location of the gland. Conclusions Type II 5α reductase plays an important role in the development and progression of benign prostatic hyperplasia. However, the expression of type Ⅰ reductase in benign prostatic hyperplasia needs further study.
