目的:建立聚乙二醇-2000(PEG)-(NH4)2SO4双水相体系萃取光度法测定亚甲基蓝(MB)的新方法。方法:测定了PEG相中亚甲基蓝吸收光谱和荧光光谱;研究了溶液酸度、盐用量、PEG用量及共存物质对体系测定的影响。结果:在溶液酸度pH7、PEG用量5.0 ml、硫酸铵加入量2.0 g、测定波长669 nm时,PEG相中MB有最大吸光度。线性回归方程Y=0.01929+0.11261X,相关系数r=0.9995,线性范围为0.08~1.50μg/ml。结论:方法具有快速、灵敏、简便的优点,用于天然水和针剂亚甲基蓝测定取得较好结果。
Objective: To establish a new extraction method for the determination of methylene blue (MB) by polyethylene glycol (PEG) - (NH4) 2SO4 aqueous two - phase system. Methods: The absorption and fluorescence spectra of methylene blue in PEG phase were measured. The effects of solution acidity, salt dosage, PEG dosage and coexisting substances on the system determination were studied. Results: The maximum absorbance of MB in PEG phase was obtained when the solution pH was 7, the amount of PEG was 5.0 ml, the amount of ammonium sulfate was 2.0 g, and the wavelength was 669 nm. The linear regression equation Y = 0.01929 + 0.11261X, the correlation coefficient r = 0.9995, the linear range of 0.08 ~ 1.50μg / ml. Conclusion: The method has the advantages of fast, sensitive and simple, and it can be used in the determination of methylene blue in natural water and injections.
