目的:通过观测姜黄素对佐剂性关节炎(AA)大鼠血清肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-β)和免疫器官重量的影响,探讨姜黄素治疗类风湿性关节炎(RA)的可能机制,为姜黄素治疗RA在临床上的运用提供理论和实验依据。方法:采用弗氏完全佐剂(FCA)复制出大鼠AA模型,将SD大鼠随机分成正常对照组、模型组、姜黄素组。采用姜黄素腹腔注射的方法进行治疗。15天后取血液,离心取血清,-20℃冰箱保存备用,采用酶联免疫法(ELISA)检测TNF-α、IL-1β的水平。取胸腺、脾脏,电子天平称重,计算重量。结果:佐剂性关节炎大鼠模型组血清TNF-α、IL-1β水平均高于正常对照组(P<0.05);经姜黄素治疗后,姜黄素组的TNF-α、IL-1β水平低于模型组(P<0.05)。姜黄素组胸腺重量较模型组低且差异有统计学意义(P<0.05)。结论:姜黄素具有抑制AA大鼠TNF-α、IL-1β的异常分泌的作用,并可以降低AA大鼠胸腺重量,从而发挥抗炎及免疫抑制作用。
Objective: To observe the effects of curcumin on the serum levels of tumor necrosis factor-α (TNF-α), interleukin-1β (IL-β) and immune organs in adjuvant arthritis (AA) The possible mechanism of arthritis (RA) provides theoretical and experimental basis for the clinical application of curcumin in the treatment of RA. Methods: Rat AA model was established by Freund’s complete adjuvant (FCA). SD rats were randomly divided into normal control group, model group and curcumin group. Curcumin intraperitoneal injection method for treatment. After 15 days, the blood was taken out and the serum was centrifuged and stored at -20 ℃ for later use. The levels of TNF-α and IL-1β were detected by enzyme-linked immunosorbent assay (ELISA). Take thymus, spleen, weighing electronic scales, calculate the weight. Results: The levels of TNF-α and IL-1β in serum of adjuvant arthritis model group were higher than those of normal control group (P <0.05). After curcumin treatment, the levels of TNF-α and IL-1β in curcumin group Lower than model group (P <0.05). The thymus weight in the curcumin group was lower than that in the model group and the difference was statistically significant (P <0.05). CONCLUSION: Curcumin can inhibit the abnormal secretion of TNF-α and IL-1β in AA rats and can reduce the weight of thymus in AA rats and thus exert anti-inflammatory and immunosuppressive effects.
