目的应用分光光度法测定炭疽疫苗的浓度。方法确定分光光度法测定炭疽疫苗的测定波长及线性范围,并对样品稳定性及方法的重复性进行分析。结果确定600nm作为检测波长;炭疽疫苗浓度在(0.25~1.25)×108个/ml之间时,A600值与菌液浓度之间具有良好的相关性(r=0.9991,自由度=3,P<0.01);样品经甲醛处理后稳定性增强;该方法具有良好的重复性。结论分光光度法可用于炭疽疫苗浓度的测定,可替代比浊法或作为炭疽疫苗浓度测定的确证方法。
Objective To determine the anthrax vaccine concentration by spectrophotometry. Methods The determination wavelength and linear range of anthrax vaccine were determined by spectrophotometry, and the stability of the sample and the repeatability of the method were analyzed. The results showed that 600 nm was the detection wavelength. There was a good correlation between the A600 value and the bacterial concentration when the anthrax vaccine concentration was between (0.25-1.25) × 108 cells / ml (r = 0.9991, 3 degrees of freedom, P < 0.01). The stability of samples after formaldehyde treatment was enhanced. The method has good repeatability. Conclusion Spectrophotometry can be used for anthrax vaccine concentration determination, and can be used as an alternative to the turbidimetric method or as a confirmatory method for anthrax vaccine concentration determination.
