目的研究假酸浆Nicandra physaloides果中的黄酮类化学成分及其免疫活性。方法采用硅胶、ODS及HPLC等色谱方法分离化合物,利用NMR、MS波谱学方法鉴定其结构,并采用脂多糖(LPS)诱导的小鼠脾淋巴细胞增殖模型评价化合物的免疫活性。结果从假酸浆果70%乙醇回流提取物的正丁醇部位分离得到13个化合物,分别鉴定为3’’-hydroxy-puerarin(1)、puerarin-6″-O-glucoside(2)、daidzein 8-C-[α-D-apiofranosyl-(1→6)]-β-D-glucopyranoside(3)、葛根素(4)、3’-methoxyl-puerarin(5)、6-C-α-L-arabinosyl-8-C-β-D-glycosylchrysin(6)、oroxylin A-7-O-β-D-glucuronide methyl ester(7)、大豆黄苷(8)、黄芩苷(9)、quercetin-3-O-β-D-galactopyranoside(10)、quercetin 3-rutinoside(11)、kaempferol 3-rutinoside(12)和quercetin 3-glucoside(13);化合物对LPS刺激的小鼠脾淋巴细胞增殖影响的实验表明,化合物1~3、7、9、10、12、13显示出对小鼠脾淋巴细胞增殖的抑制作用,而化合物6表现出对小鼠脾淋巴细胞增殖的促进作用。结论化合物1~7为首次从茄科植物中分离得到,8~13为首次从该属植物中分离得到,且化合物1~3、6、7、9、10、12、13可能具有潜在的免疫活性。
Aim To study the flavonoid constituents and immunological activities of Nicandra physaloides fruit. Methods The compounds were isolated by chromatography on silica gel, ODS and HPLC. Their structures were identified by NMR and MS spectroscopy. The immunological activity of the compounds was evaluated by lipopolysaccharide (LPS) -induced mouse spleen lymphocyte proliferation model. Results Thirteen compounds were isolated from the n - butanol fraction of 70% ethanol reflux extract of pseudo - acid berry and identified as 3 ’- hydroxy-puerarin (1), puerarin-6 "-O-glucoside (4), 3’-methoxyl-puerarin (5), 6-C-α-L-glucopyranoside (3) arabinosyl-8-C-β-D-glycosylchrysin (6), oroxylin A-7-O-β-D- glucuronide methyl ester (7), daidzein (8), baicalin (9), quercetin- Experiments on the effects of compounds on the proliferation of splenic lymphocytes stimulated by LPS in mice by O-β-D-galactopyranoside (10), quercetin 3-rutinoside (11), kaempferol 3-rutinoside (12) and quercetin 3-glucoside , Compounds 1 to 3, 7, 9, 10, 12, and 13 showed inhibitory effects on mouse spleen lymphocyte proliferation, while Compound 6 showed a promoting effect on mouse splenic lymphocyte proliferation.Conclusion Compounds 1 to 7 are the first Times from the Solanaceae plants isolated, 8 to 13 for the first time isolated from the genus plants, and compounds 1-3,6,7,9,10,12,13 may have potential immunocompetence.
