目的为了探讨姜黄素及其类似物H8治疗糖尿病肾病小鼠的作用及机制。方法建立糖尿病肾病小鼠模型,用5mg·kg-1姜黄素及其类似物H8灌胃治疗小鼠6周,HE染色观察肾病理形态学变化。实时荧光定量PCR检测小鼠肾组织中白介素6(IL-6)与核转录因子(NF-κB)mRNA表达;Western蛋白质印迹法检测小鼠肾组织中IL-6蛋白表达;免疫荧光方法检测各组小鼠肾组织中NF-κB蛋白表达。结果姜黄素及其类似物H8能明显降低糖尿病肾病小鼠体质量、血糖、血脂(P<0.01,P<0.05);HE染色结果显示肾组织炎症侵袭减轻。姜黄素及其类似物H8可显著降低尿素(UR)、肌酐(CR)、尿酸(UA)(P<0.01,P<0.05);经过H8治疗,糖尿病肾病小鼠肾组织中l L-6,NF-κB mRNA显著降低(P<0.01,P<0.05),并且IL-6和NF-κB蛋白表达明显下降(P<0.01)。结论姜黄素及其类似物H8通过抑制炎症因子基因和蛋白的表达,治疗糖尿病肾病小鼠。
Objective To investigate the effect of curcumin and its analogue H8 on diabetic nephropathy in mice and its mechanism. Methods The mouse model of diabetic nephropathy was established. The mice were treated with 5 mg · kg -1 curcumin and its analogue H8 for 6 weeks. The pathological changes of the kidney were observed by HE staining. The expression of interleukin-6 (IL-6) and nuclear transcription factor (NF-κB) mRNA were detected by real-time fluorescence quantitative PCR in renal tissues of mice; Western blotting was used to detect the expression of IL-6 protein in the kidney of mice; The expression of NF-κB in renal tissue of mice. Results The curcumin and its analogue H8 significantly reduced the body weight, blood glucose and blood lipid of diabetic nephropathy mice (P <0.01, P <0.05). The results of HE staining showed that the inflammation and invasion of renal tissue were alleviated. Curcumin and its analogues H8 can significantly reduce the levels of urinary (UR), creatinine (CR) and uric acid (UA) (P0.01, P0.05). After H8 treatment, the levels of IL- NF-κB mRNA was significantly decreased (P <0.01, P <0.05), and the expression of IL-6 and NF-κB was significantly decreased (P <0.01). Conclusion Curcumin and its analogue H8 can treat diabetic nephropathy mice by inhibiting the expression of inflammatory cytokines and proteins.
